Penfolds' winemaking centre is at Nuriootpa in Barossa Valley, and there is also a small working winery, built at the turn of the century, at Magill .

Penfolds owns about 500 hectares (1200 acres) of vineyards in South Australia's top winegrowing areas: the Barossa Valley, the Clare Valley, the Eden Valley, Adelaide, McLaren Vale and Coonawarra. It also buys grapes from growers in Langhorne Creek, Padthaway and the Adelaide Hills.

The company's present-day reputation rests on its skills in blending grapes from these regions to make a range of red, white and fortified wines each of which maintains a distinctive style from vintage to vintage. With its red wines, Penfolds is noted for skilful handling of oak maturation, and especially the use of American oak as a partner for Shiraz (Syrah).
